Occurrence of methylated purine bases in yeast ribonucleic acid.

چکیده
A number of methylated purine bases have recently been reported to occur in small quantities in the urine of normal human subjects. These include 1-methylguanine, iV*-methylguanine (6-hydroxy-2-methylaminopurine) ,I 1-methylhypoxanthine, and 8-hydroxy-7-methylguanine (1) in addition to 7-methylguanine, previously known as a urinary constituent (2). These compounds were found to be metabolites of endogenous origin (3). Their metabolic role has not been elucidated. The present study was undertaken to determine whether some of these methylated purines may occur as minor components of nucleic acids. Because of their ready availability, preparations of yeast ribonucleic acid and deoxyribonucleic acids from calf thymus and herring sperm were first examined.

A number of additional purines and pyrimidines have recently been described as minor constituents of ribonucleic acids. Cohn (1957, 1958, 1959) and Davis & Allen (1957) have described an unusual pyrimidine nucleotide from yeast ribonucleic acid, thymine, 2-methyladenine, 6-methylaninopurine and 6-dimethylaminopurine have been identified as components ofribonucleic acid from several sources (Lit...
متن کاملThe occurrence and distribution of thymine and three methylated-adenine bases in ribonucleic acids from several sources.
It has been known for several years that deoxyribonucleic acid may normally contain 5-methylcytosine (Wyatt, 1951), 5-hydroxymethylcytosine (Wyatt & Cohen, 1953), and 6-methylaminopurine (Dunn & Smith, 1955, 1958a) in addition to adenine, guanine, cytosine and thymine. The first evidence of such additional bases naturally present in ribonucleic acid is the recent isolation of an unidentified nu...
متن کاملThe purine and pyrimidine composition of yeast ribonucleic acid.
Various chromatographic, spectrophotometric, and ion exchange procedures have been used recently with somewhat varying results for the analysis of yeast ribonucleic acid for its constituent purine and pyrimidine components (l-5). The reasons for the variations may be due in part to differences in the samples analyzed and in part to deficiencies in the methods used. Some of these deficiencies ha...
